Test Principale: This assay employs a two-site sandwich ELISA to quantitate CCR5 in samples. An antibody specific for CCR5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCCR5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCR5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCR5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: CCR5 is a member of the beta chemokine receptors family of integral membrane proteins.In Humans, the CCR5 gene location is on the short (p) arm at position 21 on chromosome 3. Certain populations have inherited the Delta 32 mutation resulting in the Genetic deletion of the CCR5 gene. Homozygous carriers of this mutation are resistant to HIV-1 infection. The CCR5 protein is a seven transmembrane protein which functions as a chemokine receptor in the CC chemokine group. The natural chemokine ligands that bind to this receptor are RANTES, MIP-1α and MIP-1β. CCR5 is predominantly expressed on T cells, macrophages, dendritic cells and microglia. It is likely that CCR5 plays a role in inflammatory responses to infection, though its exact role in normal immune function is unclear.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
