Description

CD16a/FCGR3A is a low affinity receptor for the Fc region of IgG. The two genes for human FCGR3A encode for CD16A and CD16B. CD16a is a transmembrane protein found on the surface of natural killer cells, activated monocytes and macrophages and a subset of T cells. CD16a is involved in phagocytosis, secretion of enzymes and inflammatory mediators, antibody dependent cytotoxicity and clearance of immune complexes. Aberrant expression or mutations of CD16a can cause susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.

Biological activity: Activity of CD16a was measured by its binding ability in a functional ELISA. Immobilized human CD16a at a concentration of 10 ug/ml (100 ul/well) can bind biotinylated human IgG1.

Molecular Weight: The recombinant human CD16a consists of 203 amino acids and has a predicted molecular mass of 23.3 kDa. The protein migrates as a band at ~45.2 kDa due to glycosylation, in SDS-PAGE under reducing conditions.

Physical form description: Lyophilized from sterile PBS, pH 7.4, 5% trehalose, 5% mannitol and 0.01% Tween-80.

Reconstitution Instructions: Reconstitute in sterile water to a concentration of 250 ug/ml by adding 200 ul water to 50 ug of protein.

Amino acid sequence: The DNA sequence encoding the extracellular domain (Met 1-Gln 208) of human CD16a (P08637-1) precursor was expressed with a C-terminal polyhistidine tag. The predicted N terminal is Gly 17.

Handling: Centrifuge the vial prior to opening.
