Application: This Human Late cornified envelope-like proline-rich protein 1 (LELP1) ELISA Kit employs a two-site sandwich ELISA to quantitate LELP1 in samples. An antibody specific for LELP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLELP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LELP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LELP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.
• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: By searching for genes in a region of chromosome 1q21 associated with atopic dermatitis (ATOD2), Sharma et al. (2007) identified LELP1. By genomic sequence analysis, Sharma et al. (2007) mapped the LELP1 gene to chromosome 1q21. Interestingly, locus 5 is 6.2 kb upstream of a late cornified envelope-like proline-rich 1 (LELP1) gene which encodes a novel small proline rich protein. a significant association of rs7534334 (tagged SNP from LELP1) SNP with log10 serum IgE levels in the patient group (P=0.0029) . a chromosomal region in close proximity to a novel gene and highlight the need for intense research on LELP1 and other genes close by with respect to atopic disorders.
