Application: This Human MAP kinase-activated protein kinase 5 (MAPKAPK5) ELISA Kit employs a two-site sandwich ELISA to quantitate MAPKAPK5 in samples. An antibody specific for MAPKAPK5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAPKAPK5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAPKAPK5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAPKAPK5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.
• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: MAP kinase-activated protein kinase 5 is a member of the serine/threonine kinase family. In response to cellular stress and proinflammatory cytokines, this kinase is activated through its phosphorylation by MAP kinases including MAPK1/ERK, MAPK14/p38-alpha, and MAPK11/p38-beta. In vitro, this kinase phosphorylates heat shock protein HSP27 at its physiologically relevant sites. Two alternately spliced transcript variants of this gene encoding distinct isoforms have been reported.MAPKAPK5 encodes a predicted 471-amino acid protein that shares 20 to 30% sequence identity with RSK, MNK, and MAPKAPK kinases. MAPKAPK5 contains the conserved protein kinase domains I through XI, which are characteristic of all protein kinases.
