Application: This Human Ribonucleases P/MRP protein subunit POP1 (POP1) ELISA Kit employs a two-site sandwich ELISA to quantitate POP1 in samples. An antibody specific for POP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPOP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for POP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of POP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Allow all reagents to warm to room temperature for at least 30 minutes before opening.
• Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.
• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: POP1 is a protein subunit of two different small nucleolar ribonucleoprotein complexes: the endoribonuclease for mitochondrial RNA processing complex and the ribonuclease P complex. This protein is a ribonuclease that localizes to the nucleus and functions in pre-RNA processing.Human POP1 encodes a predicted 1, 024-amino acid protein that has a pI of 9.86. On Northern blots, human POP1 is expressed as a major 4.1-kb and minor 6-kb transcript, perhaps due to alternative polyadenylation signals. Anti-POP1 antibodies recognize a doublet of approximately 115 kD on Western blots. In HeLa cells, human POP1 localizes to the nucleus and strongly accumulates in the nucleolus. In co-immunoprecipitation studies, POP1 was associated with both RNase P and RNase MRP snRNAs, and with RNase P enzymatic activity.
