Test Principale: This assay employs a two-site sandwich ELISA to quantitate TLL2 in samples. An antibody specific for TLL2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTLL2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TLL2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TLL2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: TLL2 encodes an astacin-like zinc-dependent metalloprotease and is a subfamily member of the metzincin family. Unlike other family members, a similar protein in mice does not cleave procollagen C-propeptides or chordin.TLL2 shares 80.3% amino acid sequence identity with BMP1, including 870 overlapping residues. Analysis by RT-PCR ELISA demonstrated weak expression of TLL2 in most tissues and moderate expression in heart and brain.TLL2 encodes a deduced 1, 015-amino acid protein with a molecular mass of 120 kD by SDS/PAGE.Whereas BMP1, TLD, and TLL1 specifically process procollagen C-propeptides at the physiologically relevant site, TLL2 lacks this activity. BMP1 and TLL1 cleave chordin, but TLL2 and TLD do not.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
