BACKGROUND: NF-kB is sequestered in the cytoplasm by IkBa, unable to translocate to the nucleus. IkB is part of the canonical pathway for NF-kB activation. IKKb is part of the IkB kinase complex, along with a second catalytic subunit, IKKa, and a regulatory subunit, IKKg. Along with IKKa, IKKb phosphorylates IkBa leading to its degradation and release of inhibition of NF-kB. The RELA subunit of active NF-kB is then free to enter the nucleus and initiate a number of signaling cascades.
