Kelch-like ECH-associated protein 1 (KEAP1) is a redox-sensitive protein that belongs to the family of BTB proteins. KEAP1 contains cysteine residues whose thiol groups play a significant role in binding KEAP1 with CUL3 protein and forms a Cullin-RING E3 ligase complex required for Nrf2 degradation. Under oxidative stress conditions, electrophiles and reactive oxygen species (ROS) modulate the thiol groups on KEAP1, thus preventing the binding with CUL3. As a result, NRF2 translocates to the nucleus and binds to antioxidant responsive elements (AREs), and activates the expression of antioxidant proteins. Activation of NRF2 has been demonstrated to be beneficial in diabetes, Parkinson’s, and Alzheimer’s’ disease. Mutations in KEAP1 are associated with solid tumors. ’s KEAP1 (Human) ELISA kit quantitatively measures KEAP1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of KEAP1 in the sample or standard.
