MMP-13 plays a role in the degradation of extracellular matrix proteins including fibrillar collagen, fibronectin, TNC and ACAN. It can Cleave triple helical collagens, including type I, type II and type III collagen, but has the highest activity with soluble type II collagen. Transcriptional regulation of MMP-13 is tightly controlled due to its potent proteolytic capacity. MMP-13 plays an important role in keratinocyte migration during wound healing, cell migration and in tumor cell invasion. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of MMP-13 in the sample.
