Test Principale: This assay employs a two-site sandwich ELISA to quantitate SPO11 in samples. An antibody specific for SPO11 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySPO11 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPO11 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPO11 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: SPO11 is similar in sequence and conserved features to the yeast meiotic recombination protein. The encoded protein belongs to the TOP6A protein family. Several transcript variants encoding different isoforms have been found for this gene, but the full-length nature of only two of them have been described. The human and the mouse SPO11 proteins share 82% amino acid identity, but only 20 to 30% with other eukaryotic homologs such as Drosophila and C. elegans. Both the mouse and human SPO11 exons 2 and 8 are subject to alternative splicing. Northern blot analysis showed that mouse Spo11 is expressed in testis and thymus as a 1.8-kb mRNA, whereas human SPO11 is expressed in testis as a 2.0-kb transcript. RT-PCR revealed that human SPO11 is also expressed in prostate, fetal testis, thymus, and some carcinoma cell lines.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
