Test Principale: This assay employs a two-site sandwich ELISA to quantitate PRSS21 in samples. An antibody specific for PRSS21 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRSS21 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRSS21 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRSS21 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: PRSS21 encodes a cell-surface anchored serine protease, which is a member of the trypsin family of serine proteases. It is predicted to be active on peptide linkages involving the carboxyl group of lysine or arginine. The protein localizes to the cytoplasm and the plasma membrane of premeiotic testicular germ cells and it may be involved in progression of testicular tumors of germ cell origin. The deduced 314-amino acid ESP1 precursor protein has a calculated molecular mass of 33.1 kD, and the 273-amino acid active form has a calculated molecular mass of 30.6 kD. ESP1 contains an N-terminal signal peptide, a propeptide, an active site catalytic triad of conserved his, asp, and ser residues, and a hydrophobic C terminus, which suggests that ESP1 is a membrane-type serine protease.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
