Test Principale: This assay employs a two-site sandwich ELISA to quantitate TMSB10 in samples. An antibody specific for TMSB10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMSB10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TMSB10 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMSB10 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Thymosin-beta-10 is closely related in sequence to TMSB4 and is also an actin-sequestering protein. In most tissues of the rat, the same molecular weight species of TMSB10 mRNA is found; This finding, together with primer extension experiments, suggested that the 2 mRNAs are transcribed from the same gene through a combination of differential promoter utilization and alternative splicing. Both mRNAs were present in pachytene spermatocytes; only the testis-specific mRNA was detected in postmeiotic haploid spermatids. Immunohistochemical analysis showed that the protein was present in differentiating spermatids, suggesting that the testis-specific TMSB10 mRNA is translated in haploid male germ cells. thymosin-beta-10 protein synthesized in adult testis was identical in size to that synthesized in brain.
