Test Principale: This assay employs a two-site sandwich ELISA to quantitate PLAC1 in samples. An antibody specific for PLAC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPLAC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PLAC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PLAC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Cocchia et al. (2000) isolated a novel gene, which they designated PLAC1, approximately 65 kb telomeric of the HPRT gene. The mouse homolog was also found to map near Hprt on the X chromosome. Human PLAC1 is transcribed in the telomeric to centromeric direction, opposite to the orientation of HPRT, and contains 3 predicted exons. It encodes a deduced 212-amino acid protein that shares 60% sequence identity with mouse Plac1. The human and mouse proteins contain a signal peptide and a ZP3 motif. Northern blot analysis detected placenta-restricted expression of a 1.7-kb PLAC1 mRNA transcript. In situ hybridization studies in mouse detected Plac1 expression from 7.5 to 14.5 days postcoitum in ectoplacental cone, giant cells, and labyrinthine trophoblasts.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
