TIMELESS (TIM) is a core component of the circadian clock. It is necessary for generating circadian rhythms and photo-entrainment in flies. In flies, TIM and PER proteins form a heterodimer and contribute to the negative feedback loop of the circadian clock. In humans, TIM is required for the electrical oscillations output by the suprachiasmatic nucleus (SCN). Human TIM also plays a critical role in DNA replication, stabilizes replication forks by forming a complex with TIPIN, and promotes DNA repair. In mice, TIM is necessary for embryonic development. Homozygous knockout of TIM in mice is embryonically lethal, whereas heterozygous TIM knockout does not affect the circadian period. Studies have demonstrated that TIM is frequently overexpressed in cancers such as breast, colon, lung, and cervical cancer. Decreasing TIMELESS expression slows the proliferation of cancer cells. Hence, TIMELESS may serve as an efficient marker for anti-cancer therapies. ’s TIMELESS (Human) ELISA kit quantitatively measures TIMELESS in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of TIMELESS in the sample or standard.
