Transient receptor potential ankyrin 1 (TRPA1) is a calcium-permeable non-selective cation channel that belongs to the transient receptor potential (TRP) channel superfamily. TRPA1 is expressed on multiple cells such as neuronal ganglia, hair cells, cardiomyocytes, and pancreatic beta cells. One of the main functions of TRPA1 is to depolarize the plasma membrane and allow the influx of Ca2+. Another function is to detect, integrate, and initiate pain signals in the peripheral nervous system. Various stimuli activate TRPA1 at the site of injury or inflammation: they include cold/hot temperatures, pungent natural compounds (allyl isothiocyanate, formalin, allicin, cinnamaldehyde), ambient toxins (nicotine, acrolein), anesthetic agents (lidocaine), and endogenous agonists (nitric oxide, H2S). Therefore, TRPA1 is considered a “chemosensor” for the body. Missense mutation of TRPA1 is associated with a rare autosomal dominant disease called Familial Episodic Pain Syndrome (FEPS). TRPA1 also plays a significant role in neuropathy, inflammation, bronchitis, osteoarthritis, migraine, fibromyalgia, and emphysema. Hence, TRPA1 could serve as an efficient target for pain relief. ’s TRPA1 (Human) ELISA kit quantitatively measures TRPA1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of TRPA1 in the sample or standard.
