Background: The human cell line blot is designed for proteomics research in screening for antibody and protein expression. It carries 8 different human cell lines with 15 µg of total cellular protein in each lane. Proteins were separated in 4-20% SDS-PAGE gel and transferred onto nitrocellulose membrane. The cell lines included in the blot are Daudi (B lymphoblast; Burkkit's lymphoma) , Raji (B lymphoblast; Burkkit's lymphoma) , K-562 (bone marrow; chronic myelogenous leukmia) , U-937 (histiocytic lymphoma) , THP-1 (monocyte; acute monocytic leukmia) , HL-60 (promyeloblast; acute promyeloblastic leukmia) , Jurkat (T lymphocyte; acute T cell leukmia) , and MOLT-4 (T lymphoblast; acute lymphoblastic leukmia) .

Background: A431 lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl fluoride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 μg/ml of aprotinin, 5 μg/ml of leupeptin. Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The A431 lysate was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 50 mM DTT.
