Although, the urea cycle is responsible for most ammonia removal in the liver, glutamine synthetase is an important secondary metabolic pathway also responsible for its removal. The HepG2 cell line has been transformed with a vector containing a glutamine synthetase gene which allows this cell line to metabolism ammonia. With its high glutamine synthetase activity, the HepG2-GS cell line has the ability to metabolize ammonia from its culture media similar to primary hepatocytes. Ammonia-metabolizing cell lines have been used for research into hybrid bioartificial liver support systems, an innovative therapy that has potential to serve as a treatment for liver failure. Therefore, this immortalized cell line may serve as an ideal model for research pertaining to ammonia metabolism systems for long term use in place of primary hepatocytes.
